摘要 目的 观察牙齿发育过程中c-mycmRNA转录和蛋白表达与细胞凋亡的发生情况 ,探讨c -myc基因在牙齿发育中对细胞凋亡的可能作用。方法 利用原位末端标记法 (TUNEL)、原位杂交及免疫组织化学技术分别对SD大鼠牙胚发育不同时期c -mycmRNA、蛋白的表达及细胞凋亡情况进行检测。结果 在牙齿发育的各期成釉器及间充质细胞中均有c -mycmRNA、蛋白阳性表达 ,早期表达明显 ,钟状期的表达减弱而牙体组织形成期成釉细胞、成牙本质细胞及间充质牙乳头细胞中c -myc表达又增强。而凋亡细胞早期集中出现在细胞生长中心处 ,牙体组织形成后星网层及牙乳头细胞中多见 ,成釉细胞、成牙本质细胞中散在。结论 c -myc基因在牙齿发育过程中不仅调节细胞生长、增殖、分化和成熟 。 Objective To investigate the role of c-myc gene and apoptosis in the different stages of tooth development.Methods Apoptosis,expression of both mRNA and protein of c-myc were detected by Tdt-mediated dUTP nick end labeling (TUNEL) and in situ hybridization and immunohistochemical staining in different stages of tooth development of SD rats.Results The expression of both mRNA and protein of c-myc were obvious in the bud and cap stages,while decreased in the bell stage,then increased in ameloblast and dentinoblast during dentin formation. The expressions of c-myc were stronger in mesenchymal cells than in epithelial cells.The expression and distribution of c-myc mRNA was consistent with that of c-myc protein. The apoptosis predominantly appeared at proliferative growth center in the earlier stage,apoptosis was detected more in the stellate reticulum cells of enamel organ and dental papilla when dentin formation.Conclusion c-myc is of great importance in the cell proliferation and differentiation and apoptosis in the tooth development.
