雌激素通过α受体亚型途径促进内皮祖细胞增殖和迁移的研究

摘要 目的观察1 7β雌二醇(雌二醇)及其选择性雌激素α受体激动剂(PPT)、β受体激动剂(DPN)对内皮祖细胞(EPC)增殖和迁移能力的影响。方法将培养的EPC分别加入不同浓度雌二醇、PPT、DPN(分别为1×10^(-10)、1×10^(-9)、1×10^(-8)、1×10^(-7)mol/L)进行培养后,分为雌二醇组、PPT组、DPN组和对照组。采用异硫氰酸荧光素标记荆豆凝集素I、DiI标记乙酰化低密度脂蛋白双染及CD1 33免疫荧光鉴定。并进行细胞增殖能力分析、EPC计数和流式细胞技术检测EPC细胞周期,Transwell技术检测EPC迁移能力的影响。结果与对照组比较,雌二醇组、PPT组吸光度值、细胞周期细胞比例、EPC迁移数目明显升高(P<0.05);与DPN组比较,雌二醇组和PPT组吸光度值、细胞周期细胞比例、EPC迁移数目明显升高(P<0.05);与DPN组(2.21±0.99)×10~5个EPC比较,雌二醇组(2.83±0.29)×10~5个和PPT组(2.56±0.11)×10~6个明显增多(P<0.05)。结论雌二醇主要通过α受体亚型途径刺激EPC的增殖和迁移。 Objective To study the effect of 17β estradial and its selective a-protagonist(PPT)and β-protagonist(DPN) on proliferation and migration of endothelial progenitor cells(EPC). Methods EPC, cultured with estradial,PPT and DPN at different concentrations(1 ×10^-10 mol/L, 1 × 10^-9 mol/L,1 × 10^-8 mol/L,1 × 10^-7 mol/L) ,were divided into estradial group,PPT group,DPN group, and identified with thiocyanic acid immunofluorescin-labelled lectin-1 staining, Dil-labelled acetylated LDL double staining and CDβ3 immunofluorescence staining. Proliferation of EPC was analyzed, EPC were calculted and cell cycle of EPC was detected by flow cytometry, migration of EPC was observed with the transwell technique. Results The light absorption value,the proportion of EPC in different cell cycles, and the number of migrated EPC were significantly higher in estradial group and PPT group than in control group and DPN group(P〈0.05). Conclusion Eestradial promotes the proliferation and migration of EPC via a-receptor subtypes.
