摘要
目的探讨环氧化酶2(cyclooxy genase,COX-2)、胸腺间质淋巴细胞生成素(thymic stromal lymphopoietin,TSLP)在雌激素化小鼠念珠菌性阴道炎模型中的表达及其在致病中的作用。方法将60只小鼠随机分为雌激素化白念珠菌感染组(EI组)、雌激素化非感染组(E组)及正常对照组(C组),EI组采用皮下注射雌激素及阴道内接种白念珠菌造模,E组仅皮下注射雌激素。念珠菌接种后第2,4,7,14天,3组小鼠阴道冲洗液中真菌量的动态变化采用集落形成单位(CFU)计数。各组小鼠阴道组织中的念珠菌感染情况,COX-2和TSLP的表达分别采用PAS染色、免疫荧光激光共聚焦检测,并进行统计学分析。结果 EI组CFU自接种后第2天升高、第4天达到高峰、之后下降。EI组CFU平均值(3.47±1.82)×103/mL高于E,C组(E,C组无念珠菌生长,P<0.01);第7天,EI组COX-2绿色荧光程度较强(0.252±0.020),明显高于E组(0.155±0.005)和C组(0.098±0.006),经比较差异具有统计学意义(P<0.001);第7天,EI组TSLP红色荧光最强(0.391±0.011)明显高于E组(0.243±0.020)和C组(0.155±0.006),经比较差异有统计学意义(P<0.001)。结论在雌激素化小鼠念珠菌感染模型中,黏膜中COX-2,TSLP表达的上调可能在念珠菌致病中起重要作用。
Objective To investigate the expression and pathological role of COX-2, and TSLP in the estrogenized mouse vaginal-candidiasis model. Methods The 60 mice were randomly divided into estrogenized Candida albicans-infected group (EI) , estrogenized non-infected group (E), and normal control group (C). Estrogenized Candida albicans-infected model was established by subcutaneous injection of estrogen and vaginal inoculation of C. albicans. While E group were only subcutaneously injected with estrogen. Dynamic changes in the amount of fungus were obtained from vaginal eluate of the 3 groups at different time points ( d2, d4, d7 and d14) after inoculation of C. albicans were detected in colony forming units (CFU). In murine vaginal tissue of each group, Candida infection, expression of COX-2, TSLP were detected with PAS staining and confocal immunofluorescence, respectively, and performed statistical analysis. Results COX-2 in the vaginal tissues of EI, E groups were significantly higher than that in C group. In the three groups, TSLP ex- pressed the highest in the EI group, followed by E group, with C the lowest (P 〈 0.01 ). Conclusion In estrogenized Candida albicans-infected mouse model, estrogen-induced upregulation of COX-2, TSLP expression in vaginal mucosa may play an important role in Candida pathogenicity.
