摘要
目的探讨IL-1β、TNF-α在实验性矽肺大鼠的表达及辛伐他汀的干预对其影响。方法本文取雄性Wistar大鼠40只随即分为对照组,模型组、干预1组,干预2组,每组10只,采用逆转录-聚合酶链反应检测其大鼠肺组织中的IL-1β和TNF-αmRNA表达水平。结果 IL-1β、TNF-α表达在模型组以及干预组在干预前表达水平无显著差异,但高于对照组(P均<0.05)。干预组在干预后IL-1β、TNF-α表达水平明显低于干预前、均低于模型组(P均<0.05),且TNF-α表达与辛伐他汀呈明显负相关。结论 IL-1β、TNF-α高表达在矽肺肺纤维化具有重要作用,辛伐他汀可以通过抑制TNF-α表达,发挥其抗纤维化作用。
Objective To study the changes of IL-1β,TNF-α levels in experimental rats with pneumoconiosis,and the changes after simvastatin treatment.Methods The rats were randomly divided into four groups,the control group,the model group rats,the first experiment group,and the second experiment group of,with 10 rats in each group;The mRNA levels of IL-1β,TNF-αwere evaluated by reverse transcription-polymerase chain reaction(RT-PCR).Results The levels of IL-1β,TNF-α in model group and the experiment group before intervention have no differences,but all were higher than those in the control groups.The levels of IL-1β and TNF-α in experiment group were significantly lower after intervention(P0.05).The expression of TNF-α had a positive relationship with simvastatin(P0.05).Conclusions The increased expression of IL-1β,TNF-α playes an important role in pulmonary fibrosis of pneumoconiosis rats,and simvastatin may inhibit pulmonary fibrosis by means of inhibitting the activity of TNF-α.
