摘要
为分析黑龙江地区28株野生黑木耳菌株的遗传多样性,并构建种质分子身份证。利用SSR分子标记对供试菌种进行遗传多样性分析,用NTSYS软件进行聚类分析并用IDAnalysis1.0软件种质分子身份证。结果表明:7对引物共检测到131个多态性片段,品种间特异性指数介于23.015~53.827之间,平均为30.00;在相似水平在0.58时,28个供试黑木耳菌株被分为3个组群。结果仅需5对引物即可将28份参试菌株完全区分开。
Genetic diversity of wild Auricularia auricula strains in Heilongjiang Province was analyzed, and a set of molecular ID were established for them. 28 wild Auricularia auricula strains were collected from Heilongjiang Province and analyzed its genetic diversity by using SSR markers through clustering method by NTSYS, and according to fragment size of allele variation, the data numeralized from the AGE bands were analyzed by the software ID Analysis 1.0. The results showed that 131 polymorphic alleles were detected with 7 SSR primers. There existed much difference in specific indices of varieties, ranging from 23.015 to 53.827 with an average of 30.00. The results from the clustering analysis by NTSYS showed that these wild strains could be divided into three groups at similar level of 0.58. 5 primers could be used to identify all 28 wild Auricularia auricula strains. The AGE pattern of each cultivar amplified with the 5 primers could be used as a molecular ID of the strains.
