摘要
目的:建立HPLC-ELSD法测定参贝北瓜颗粒中贝母素甲和贝母素乙的含量。方法:采用AglientC18柱(150mm×4.6mm,5μm)为色谱柱;流动相为乙腈∶1%二乙胺溶液(70:30);流速1.0mL.min-1;柱温35℃;ELSD800蒸发光检测器:漂移管温度:40℃,载气压力2.5MPa。结果:贝母素甲在0.249~1.492μg/mL范围内呈良好的线性关系(r=0.9991),平均回收率为98.6%,RSD为2.54%;贝母素乙在0.246~1.476μg/mL范围内呈良好的线性关系(r=0.9995),平均回收率为99.4%,RSD为2.10%。结论:该法准确可靠,可作为该制剂质量控制方法。
Objective:To esablish a HPLC-ELSD method for determining the content of peimine and peiminine in shenbei beigua keli. Methods: a Agilent C18 column (150min × 4. 6mm, 5um) was used. The mobile phase was acetonitrile -1 % diethylamine(70 : 30) w/th a flow rate of 1.0mL ·min -1. An evaporate light-scattering detector(ELSD) was used as detector with gas pressure was 2.5MPa and drift tube temperature of 40 ℃. Results:The lingear range of peimine was 0. 249 - 1. 492 ug/mL(r= 0. 9991). The average recovery was 98.6% , RSD was 2.54 %. The lingear range of peiminine was 0. 246- 1. 476 ug/mL(r=0. 9995). The average recovery was 99.4 %, RSD was 2. 10%. Conclusion: The method isreliable , accurate and can be applied to the quality control of the preparetion.
