摘要
目的:探索胰岛素抵抗的卵泡膜细胞对腺苷酸环化酶激活剂(Forskolin)的反应变化以及中药隐丹参酮对卵泡膜细胞甾体激素合成的调控作用。方法:采用猪卵巢卵泡膜细胞进行体外培养,应用渥漫青霉素(Wortmannin)诱导构建胰岛素抵抗的细胞模型,并应用隐丹参酮和Forskolin处理各组细胞,检测中药隐丹参酮和/或Forskolin作用后各组的培养液葡萄糖浓度和激素水平的变化以及胰岛素信号分子和甾体激素合成关键酶的mRNA表达变化。结果:卵泡膜细胞发生胰岛素抵抗后,睾酮(T)、孕酮(P)合成能力明显增强,17α羟化酶(CYP17)、3β-羟基甾脱氢酶(3β-HSD)、过氧化物酶增殖体激活受体γ(PPARγ)、细胞色素芳香化酶(P450arom)和类固醇激素合成急性调节蛋白(STAR)的mRNA表达量增加,胰岛素抵抗卵泡膜细胞对Forskolin的生物效应反应性增强。中药隐丹参酮能够缓解卵泡膜细胞胰岛素抵抗,同时也降低了培养液中睾酮的水平和甾体激素合成酶CYP17和STAR的mRNA水平。结论:中药隐丹参酮能够降低由于卵泡膜细胞胰岛素抵抗所产生的功能异常,提示中药增敏剂可以用于多囊卵巢综合征的治疗,可以起到改善生殖功能的作用。
Objective: To investigate the reaction to Forskolin in thecal cells of insulin resistance and the effect of eryptotanshinone (Cry) on steroidogenesis Methods: The model of IR granulosa cells was established in vitro culture by treating with wortmannin and/or cryptotanshinone. The residual glucose concentration, medium hormone level and mRNA expression of key enzymes of hormone synthesis were tested. Results: The secretion of T and P was increased and the expression of CYP17, 3β -HSD, PPAR γ, P450 atom and StAR mRNA also increased, when thecal cells occur IR. The biological effects of forskolin on insulin resistance in vitro porcine thecal cell is amplified. Cryptotanshinione down-regulated the level of thecal cell insulin resistance, CYP17 and STAR mRNA expression and medium testosterone level.Conclusion: Cryptotanshinone can be used for the treatment of polycystic ovary syndrome, it also can improve the reproductive function.
