摘要 目的:探讨碱性成纤维细胞生长因子(bFGF)对骨髓基质细胞(BMSC)粘附、增殖和分化等生物学效应的影响方法:利用体视学计数、MTT法及ALP试剂盒分别测定不同浓度bFGF诱导一定时间后BMSC的粘附特性、增殖和分化情况的变化结果:10 ng/mL bFGF明显促进BMSC的粘附,但是200 ng/mL bFGF反而不利于细胞粘附;在细胞增殖和分化测定中,100 ng/mL bFGF明显促进细胞增殖,但细胞碱性磷酸酶含量也最低结论:碱性成纤维细胞生长因子对骨髓基质细胞的生物效应是复杂和多方面的,可以作用于骨髓基质细胞的粘附、增殖和分化等多个环节。 Aim: To investigate the biological effects of basic fibroblast gr owth factor(bFGF) on the cell adhension, proliferation and differentiation of bo ne marrow stromal cells(BMSC). Methods: After induction of different concentrati ons of bFGF, calculate the attached cells with stereological method, measure cel l proliferation with MTT method and cell differentiation with ALP measurement ki Results: 10 ng/mL bFGF can promote cell adhension significantly, while 200 ng/mL bFGF may inhibit cell attachement; we also found that 100 ng/mL bFGF can promote cell proliferation significantly, however ALP was the lowest. Conclusion: bFGF may play compound and multiple roles in the biological effect of BMSC, including cell adhension, proliferation and differentiation. This effect is connected with the concentration of bFGF
