摘要
目的】 探讨孕期饮酒致髓鞘发育异常机制。【方法】 以3H TdR掺入体外培养少突胶质细胞作为细胞增殖指标。观察酒精及其代谢产物乙醛对细胞增殖的影响。【结果】 体外处理 2 4h和 48h后 ,绝大多数剂量组CPM值均低于相应时间的对照组 (P <0 0 5 ) ,且随酒精、乙醛浓度增加 ,细胞增殖能力显著下降 ,存在剂量反应关系。CPM值与实验剂量呈明显负相关。【结论】 酒精、乙醛对少突胶质细胞生长有明显抑制作用。酒精及其代谢产物乙醛可推迟细胞增殖和诱发细胞异常变化 ,这可能是胎儿酒精综合征中的髓鞘发育异常的主要原因之一。
Objective To explore the mechanism of myelin development abnormality induced by drinking alcohol during pregnancy. Methods \ In this study,oligodendrocytes were cultivated and incorporation of 3H\|TdR in vitro was used as an indicator of cell proliferation The effect of alcohol and its metabolite acetaldehyde on proliferation was observed. Results The result showed that after treatment for 24 and 48 hours,cpm values in most of experimental groups were lower than those of the controls( P <0 05).With inceease in doses of alcohol and acetaldehyde,the ability on proliferation of oligodendrocytes was decreased with a dose\|respone relationship.The CPM values were significantly negatively related with doses. Conclusion These results suggested that alcohol and acetaldehyde have a significant restrained action to oligodendeocytes. Alcohol and its metabolite are able to delay cell proliferation and induce cell abnormal differentiation,this is probably one of the mechanisms of myelin development abnormalities in fatal alcohol syndrome.
