摘要 目的了解血浆黏附分子颗粒膜蛋白(GMP-140)水平及其在肾组织的表达与狼疮肾炎(LN)活动。肾脏病理及肾功能损害的关系。方法用酶联免疫吸附法(ELISA)测定血浆GMP-140水平,并用免疫组织化学染色法,观察正常肾和狼疮肾组织GMP-140表达。结果LN患者较正常人以及活动LN较非活动LN,血GMP-140水平显著升高;LN紧组织GMP-140表达较正常肾组织明显增强,且狼疮肾组织GMP-140+细胞数与血浆GMP-140水平和狼疮肾活动指数明显正相关,与肾脏病理和功能损害有一定相关关系。结论GMP-140在介导LN中起重要作用,其血浆水平或肾组织表达水平可作为反映LN活动和进行性肾损害的指标。 Objective To investigate the plasma level and in situ expression of granular membrane protein(GMP-140) in lupus nephritis (LN), and their correlations with the renal injuries. Methods Plasma GMP-140 was measured with ELISA in LN patients and normal control. GMP-140 expression in the kidney was measured by a microwave-based immunohistochemistry. Results There was a significant elevation of plasma GMP-140 level and marked up-regulation of GMP-140 expression in renal tissue of LN patients as compared to the normal control. The elevated level of plasma GMP-140 and its increased expression in were strongly correlated with LN activity, renal function and histological changes. Conclusion GMP-140 may play an important role in the mediation of LN pathogenesis and its plasma level and expression in renal tissue may be used as an index of the disease activity and progressive renal injuries.
