摘要 [目的]探讨利用响应面分析法(Response surface methodology,RSM)优化白花蛇舌草总黄酮的提取工艺的可行性。[方法]通过单因素试验考察液固比、提取温度、提取时间、乙醇浓度及提取次数等5个因素对白花蛇舌草总黄酮提取率的影响,并在此基础上通过中心组合试验设计和响应面分析优化白花蛇舌草总黄酮的提取工艺。[结果]当液固比为26.93、提取温度80℃、提取时间为110.21 min、乙醇浓度为77.77%时,白花蛇舌草总黄酮提取率最高,为0.94%。[结论]采用RSM法优化得到的提取条件参数准确可靠,具有实用价值。 [Objective] To optimize the extraction conditions of total flavonoids from Hedyotis diffsua Willd.based on response surface methodology.[Methods] Single factor experiments were conduced to examine the effects of the ratio of water to Hedyotis diffsua Willd.,extraction temperature,extraction time,ethanol concentration and times of extraction on the total flavonoids yield.On basis of which,center-united experimental design and response surface methodology were applied to optimize extraction conditions of total flavonoids from Hedyotis Diffsua Willd.[Results] The optimal conditions of flavonoids extraction were obtained as: the water/solid ratio of 26.93,extraction temperature of 80℃,extraction time of 110.21min,ethanol concentration of 77.77%.Under the above condition,the two-time extraction yield was up to 0.94%.[Conclusion] The extraction conditions parameters obtained based on response surface methodology were accurate,realiable and practical.
