摘要
目的探讨白芍总苷(tatol glucosides of paeony,TGP)对葡聚糖硫酸钠(dextran sulfate sodium,DSS)致大鼠实验性结肠炎的治疗作用及机制。方法由DSS诱导大鼠实验性结肠炎模型,给予白芍总苷治疗,观察大鼠一般状况、疾病活动度指数(Disease activity index,DAI)和组织学评分,ELISA法检测大鼠结肠组织TNF-α、IL-4和PGE2水平,免疫组化法检测NF-κBp65的表达水平。结果白芍总苷能明显改善结肠炎大鼠一般状况,减低DAI评分和组织学评分。与模型组相比,TNF-α、PGE2和NF-κBp65水平均显著降低(P<0.05),IL-4水平虽有上升趋势,但差异无显著性。结论白芍总苷可能通过抑制NF-κBp65活化,减少TNF-α和PGE2等炎症因子表达来有效缓解结肠炎大鼠结肠炎症状态。
Objective To study the therapeutic effects of paeony on rats' ulcerative colitis caused by dextran sulfate sodium(DSS).Methods In the experiment,the general state,the disease activity index(DAI) and the histological of rats were observed and evaluated,the level of TNF-α,IL-4 and PGE2 and the level of NF-кBp65 were measured through ELISA while the sample rats suffering from ulcerative colitis that caused DSS were treated by using tatol glucosides of paeony(TGP).Results It was obvious that TGP could greatly change the general state of rats as well as ameliorate DAI and the general histological.Compared with the model group,the level of TNF-α,PGE2 and NF-кBp65 decreased significantly(P0.05) while the expression of IL-4 was higher,which showed no significant difference.Conclusion TGP can effectively attenuate experimental ulcerative colitis through inhibiting the activity of NF-кBp65 and decreasing the level of inflammatory factor.
