摘要
目的探讨TGF-β1基因作用于平滑肌细胞从而促进内皮细胞黏附的作用。方法用单价阳离子脂质体转染试剂DOTAP转染pMAMneo TGF-β1于原代培养的平滑肌细胞,经G418筛选。检测转染pMAMneo TGF-β1和转染pMAMneo的平滑肌细胞黏附内皮细胞的情况。结果经G418筛选,转染pMAMneo TGF-β1组转化生长因子TGF-β1基因在平滑肌细胞得到有效表达,内皮黏附实验表明转染pMAMneo TGF-β1组平滑肌细胞表面有大量内皮细胞黏附[(32±2)/高倍视野],而对照组平滑肌细胞仅有少量内皮细胞黏附[(11±1)/高倍视野],其与转染pMAMneo组相比,差异显著(P<0.01)。结论 TGF-β1基因能有效作用于血管平滑肌细胞从而促进内皮细胞黏附,在血管再生中具有重要作用。
Objective To examine the effect of transforming growth factor β1(TGF-β1) gene on promoting endothelial cells adhesion to smooth muscle cells.Methods Smooth muscle cells were transfected with pMAMneo TGF-β1(TGF-β1 group) and pMAMneo(control group) by DOTAP liposome transfection reagent,respectively.The positive cell clones were selected with G418,and the expression of TGF-β1 was detected by fluorescence analysis.The number of endothelial cells adhering to the transfected smooth muscle cells were determined by microscopy.Results TGF-β1 gene was successfully expressed in the smooth muscle cells transfected with pMAMneo TGF-β1.A large number of endothelial cells adhered to the surfaces of the smooth muscle cells of the TGF-β1 group,which was significantly greater than that of the control group [(32±2) vs(11±1),P〈0.01].Conclusion TGF-β1 gene can induce endothelial cells adhesion to smooth muscle cells,which is helpful for vascular tissue engineering.
