HPLC-ELSD method to determine encapsulating efficiency and loading capacity of ginkgolide B in PEG-PLGA nanoparticles

摘要 目的建立PEG-PLGA纳米粒中银杏内酯B(GB)的包封率和载药量测定的HPLC-ELSD方法。方法色谱条件为:Shiseido C_8柱(250 mm×4.6 mm,5μm);V(甲醇):V(四氢呋喃):V(水)=31:11:58为流动相;蒸发光散射检测器检测参数:蒸发口温度105℃,载气(N_2)流速1.2 L·min^(-1),柱温45℃。应用低温超速离心法和凝胶过滤相结合的方法纯化纳米粒。结果本次建立的HPLC-ELSD方法在GB质量浓度50~500 mg·L^(-1)内线性关系良好,r=0.999 1;重复性试验RSD为4.28%;平均回收率为95%~105%;载GB的PEG-PLGA纳米粒中杂质对GB的测定无干扰。GB的包封率为75.33%,载药量为3.77%。结论该方法简单、快速、准确,可用于载GB的PEG-PLGA纳米粒中GB包封率和载药量的测定。 A/M To establish a HPLC method with evaporative light scattering detector (ELSD) to determine the encapsulating efficiency and loading capacity of ginkgolide B(GB) in poly(ethyleneglycol) -poly( L-lactic-co-glycolic acid) (PEG-PLGA) nanoparticles. METHODS Chromatographic conditions were as follows: Shiseido C8 column (250 mm ×4.6 mm, 5 μn) ; V( methanol) : V( tetrahydrofuran ): V(water ) = 31 : 11:58 as mobile phase. ELSD parameters were set as follows: evaporation temperature was 105℃, carrier gas (N2) flow rate was 1.2 L·min-1 and column temperature was 45℃. Low temperature ultracentrifugation and gel filtration methods were used to purify nanoparticles. RESULTS In the range of 50 - 500 mg·L^-1, all plots were linear and the correlation coefficients were high than 0.999 0; the relative standard deviation (RSD) of the reproducibility was 4.28 % ; average recovery was in the range of 95 % - 105 %. As the dosage of GB was 1 mg, the encapsulating efficiency(EE) and the loading capacity(LC) was 75.33% and 3.77%, respectively. CONCLUSION The method is simple, rapid, accurate and can be used for quality control of GB loaded nanopartieles.
