摘要
目的探讨河南新乡地区人类8-羟基鸟嘌呤糖苷酶(hOGG1)基因Ser326Cys单核苷酸多态性与急性心肌梗死(AMI)的关系。方法用病例-对照研究设计,选取AMI患者(观察组)和与之年龄、性别匹配且行冠状动脉造影显示冠状动脉正常的对照组各216例,运用聚合酶链式反应-限制性片段长度多态性技术检测hOGG1基因Ser326Cys单核苷酸多态性,用全自动生物化学分析仪测定总胆固醇(TC)、三酰甘油(TG)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)、空腹血糖(FBG)、血尿素氮(BUN)和肌酐(Cr)。结果观察组TC、LDL-C和FBG水平较对照组明显增高(P<0.05);观察组Cys/Cys基因型频率和Cys等位基因频率明显高于对照组(P<0.05),Ser/Cys基因型频率和Ser等位基因频率明显低于对照组(P<0.05)。与携带Ser/Ser或Ser/Cys基因型者比较,携带Cys/Cys基因型的个体患AMI的风险增加(OR=2.14,95%CI:1.64~3.77)。观察组携带Cys/Cys基因型的TC、LDL-C和FBG水平明显高于Ser/Ser+Ser/Cys基因型(P<0.05)。结论 hOGG1基因Ser326Cys单核苷酸多态性可能与AMI的发生相关。
Objective To investigate the association of human 8-hydroxyguanine glycosylase(hOGG1) Ser326Cys gene single nucleotide polymorphism and acute myocardial infarction(AMI) in Xinxiang of Henan province,China.Methods A case-control study was carried out.The hOGG1 gene Ser326Cys polymorphism was detected by polymerase chain reaction restriction fragment length polymorphism in 216 patients with AMI(observation group) and 216 age-and sex-matched controls(control group) with normal coronary artery by coronary arteriongraphy.The levels of total cholesterol(TC),triglyceride(TG),high density lipoprotein-cholesterol(HDL-C),low density lipoprotein-cholesterol(LDL-C),fasting blood glucose(FBG),blood urea nitrogen(BUN) and serum creatinine(Cr) were detected with automatic biochemistry analyzer.Results Compared with control group,the levels of TC,LDL-C and FBG in observation group were significantly higher(P〈0.05),the Cys/Cys genotypic frequency and Cys allele frequency of hOGG1 in observation group were significantly higher(P〈0.05),but the Ser/Cys genotypic frequency and Ser allele frequency in observation group were significantly lower(P〈0.05).The risk of AMI was higher in the patients with Cys/Cys genetype than that in patients with Ser/Ser or Ser/Cys genetype(OR=2.14,95%CI:1.64-3.77).In observation group,the levels of TC,LDL-C and FBG in patients with Cys/Cys genetype were significantly higher than those in patients with Ser/Ser+Ser/Cys genetype(P〈0.05).Conclusion The development of AMI may be associated with hOGG1 Ser326Cys gene single nucleotide polymorphism.
