姜油和6-姜酚对小鼠肝脏主要Ⅱ相代谢酶活性及其mRNA表达的影响 被引量:4

Effect of Ginger oil and 6-gingerol on the major liver phase Ⅱ metabolizing enzymes in vivo in mice

摘要 目的研究姜油和6-姜酚对小鼠肝脏UGT、SULT1A1和GST活性及其mRNA表达的影响。方法给小鼠灌服不同剂量的姜油和溶媒对照液,每天2次,连续3天。以分光光度法,检测肝脏中上述代谢酶活性;以RT-PCR法,检测UGT1A1/SULT1A1mRNA在肝脏中的表达。结果姜油(15 mg·kg-1)对小鼠UGT和GST活性有显著的诱导作用(P<0.05或P<0.01);以2-萘酚和4-硝基酚为底物测定SULT1A1活性时,姜油(15,3 mg·kg-1)对SULT1A1均有明显的诱导作用(P<0.05或P<0.01);RT-PCR结果显示,仅有10 mg·kg-1的6-姜酚能够提高UGT1A1/SULT1A1 mRNA的表达。结论临床用药期间,应注意食用姜类产品对Ⅱ相药物代谢酶的影响。 Objective To investigate the effect of ginger oil and 6-gingerol on liver uridine-diphosphate glucuronosyltransferases(UGT),sulfotransferase 1A1(SULT1A1) and glutathione S-transferase(GST) activities and their mRNA expressions in mice.MethodsThe mice were treated with different dosages of Ginger oil or vehicle respectively two times a day for continuous three days.The activities of UGT,SULT1A1 and GST were detected by spectrophotometry;expression of UGT1A1/SULT1A1 mRNA in liver was estimated by real-time fluorescence quantitative PCR method.ResultsCompared with control group,high-dosage(15 mg·kg^-1) of Ginger oil considerably increased UGT and GST activity(P〈0.05 orP〈0.01).High and medium-dosage(15,3 mg·kg^-1) of Ginger oil significantly elevated SULT1A1 activity(P〈0.05 or P〈0.01).RT-PCR results indicated that only 6-Gingerol(10 mg·kg^-1) can induce the expression of UGT1A1/SULT1A1 mRNA in the liver.ConclusionIt is necessary to consider the possible influence of ginger products on the activities of the phaseⅡdrug-metabolizing enzymes.
