摘要
建立小鼠顺铂耳毒性模型,研究不同剂量的顺铂对小鼠耳蜗中Calpain表达水平的影响。将70只成年BALB/c小鼠随机分成4组,用不同剂量的顺铂进行干预后,通过听脑干反应测试用药前后小鼠听力的变化;应用Western blot以及免疫荧光检测耳蜗中Calpain的表达。不同剂量顺铂组小鼠的体重和听力明显下降,随着给药时间的增加,Calpain1在耳蜗中的表达水平逐渐减弱,而Calpain2在耳蜗中的表达水平逐渐增强。应用顺铂可建立小鼠耳毒性模型;正常小鼠耳蜗中存在Calpain1和Calpain2的表达,并且calpain2与顺铂所致的耳毒性相关。
Objective Establishing a mouse model of cisplatin ototoxicity, studying the effects of cisplatin of different doses on the expression of Calpain in mouse cochlea. Methods: 70 BALB/c mice were randomly divided into five groups, after intervention with different doses of eisplatin, detecting the auditory changes before and after treatment through the auditory brainstem response tests. Western blot and immunofluorescence were applied to detect the expression of Calpain in the cochlea. Results: The weight and hearing of mice decreased significantly in the goups which treated with different doses of cisplatin. With prolongation of drug administration time, the expression of Calpainl in the cochlea decreased gradually, while the expression of Calpain 2 in the cochlea increased gradually. Conclusion: Application of cisplatin can establish the mice ototoxicity model. Normal mouse cochlea exists the expression of calpainl and calpain2, moreover, calpain2 is associated with cisplatin-induced ototoxicity.
