摘要
目的 研究视网膜母细胞瘤蛋白 (pRB)及其相关基因在脑胶质瘤中的异常表达情况。方法 用免疫组化的方法对 2 3例脑胶质瘤 (包括 14例星形细胞瘤和 9例胶质母细胞瘤 )以及 6例正常脑组织中 pRB以及cyclinD1、p16基因的表达进行检测。 结果 2 3例肿瘤中有 8例 pRB缺失(34 8% ) ,17例cyclinD1过度表达 (73 9% ) ,13例 p16缺失 (5 6 5 % ) ,三种因子的异常均与肿瘤分型有关 (P <0 0 1)。由它们组成的调控路径作为整体在肿瘤组织中异常率为 82 6 % (19/ 2 3) ,与正常脑组织 (1/ 6 ,16 7% )相比有显著性差异 (P =0 0 0 6 ) ,而星形细胞瘤 (10 / 14,71 4% )和胶质母细胞瘤(9/ 9,10 0 % )之间并无统计学差别 (P =0 12 4) ,说明此路径异常与肿瘤发生有关而与分型无关。pRB在此调控路径中处于中心位置 ,与肿瘤的恶化有关。 结论 pRB及其相关基因的异常与脑胶质瘤发生关系密切 ,可以作为探讨肿瘤病因 ,诊断和治疗的重要靶位。
Objective This experiment was to study the abnormal expression of pRB and its associated genes in gliomas.Methods The expressions of pRB and cyclin D1,p16 genes were screened in 23 gliomas (including 14 astrocytomas and 9 glioblastomas) and 6 normal brain tissues by immunohistochemical method.Results In 23 gliomas,8 cases had pRB deletion(34 8%),17 cases had cyclin D1 overexpression(73 9%),and 13 cases had p16 deletion(56 5%);Abnormalities of these proteins were closely related to tumor types ( P< 0 01).The alteration of regulatory pathway which consisted of these factors was seen in 82 6% (19/23) cases and had marked difference from normal brain tissues (1/6,16 7%)( P= 0 006).However,there was no statistical significance between astrocytomas (10/14,71 4%) and glioblastomas (9/9,100%),which indicated that the pathway abnormalities were only associated with tumorigenesis but not with tumor types.PRB gene was in the center of this pathway and related with tumor aggravation.Conclusion The abnormalities of pRB and its associated genes are closely related with tumorigenesis and they are probably important targets in studying the etiology,diagnosis and gene therapy of tumors.
