摘要
目的 :建立了反相HPLC测定人血清中咖啡因浓度的方法。方法 :甲醇 水为流动相 ,茶碱为内标 ,紫外2 54nm处检测。结果 :线性范围 2 .5~ 4 0 μg/ml,日内RSD为 1 .2 8%~ 3.78% ,日间RSD为 2 .1 8%~ 3.56% ,方法平均回收率为 99.86% ,最低检测浓度为 1 0 0ng/ml。结论 :此法具有操作简便、省时、分离度好等特点 。
OBJECTIVE:To establish a reverse phase HPLC method for the determination of caffeine in human serum.METHOD:The mobile phase was methanol water.Internal standard was theophylline.The detection wavelength was 254nm.RESULTS:The method was linear within the range of 2.5 to 40μg/ml and the limit of detction was 100ng/ml.The variations of within day ranged from 1.28% to 3.78% and those of day to day from 2.18% to 3.56%.The average recovery of caffeine was 99.86%.CONCLUSION:This assay is rapid,accurate,sensitive and simple,and can be applied to the study of the pharmacokinetics and pharmacodynamics of caffeine in clinical practise.
