链球菌表面烯醇化酶重组蛋白多克隆抗体的制备

摘要 制备M6型GAS表面烯醇化酶抗体。皮下多点注射GAS表面烯醇化酶重组蛋白(rSEN)免疫家兔,采用亲和层析纯化免疫血清,通过SDS-PAGE、ELISA、Western-Blot等方法检测抗体的特异性,并初步用制备的抗体检测了M6血清型GAS表面烯醇化酶的表达。制备的抗体蛋白纯度较高,检测的特异性较强,用制备的抗体可以检测到活体菌表面的烯醇化酶。成功制备并纯化得到了GAS表面烯醇化酶多克隆抗体。 To prepare anti - rSEN antibody and to detect the express of surface enolase of GAS. Rabbit was administered three times with rSEN subcutaneously by mudi - dot, pured immune serum by affinity chromatograph and detected the specification of anti - rSEN polyelonal antibody with SDS - PAGE, Western - Blot and ELISA. The expression of surface enolase of serotype M6 of GAS also was determined with anti - rSEN antibody. Prepared anti - rSEN polyclonal antibody was profuse and specification is obvious. The purified anti - rSEN antibody can detect the express of surface enolase of GAS. Anti - rSEN polyclonal antibody was succusfully prepared and purified.
