摘要
观察美多巴和多巴胺D_1受体拮抗剂SCH23390对大鼠体温的调节作用。实验大鼠预先分别给予两种药物后置 于相同的热环境中,观察体温升高速率。同时用放射配基结合法检测纹状体中D_1R的平衡解离常数和最大结合容量, 用放射反应试剂盒测定纹状体中。cAMP的浓度,用流式细胞仪检测纹状体细胞中钙调素(CaM)的相对活性。结果 表明,多巴胺受体激动剂美多巴能明显加快动物升温,而D_1R拮抗剂SCH223390则明显阻止升温,两种药物的作用可 能有不同的分子机制。
The effects of Madopar and SCH23390 on rats' thermoregulation were observed. Rats were put into the hot environment after been treated with Madopar or SCH23390, and the raise rate of rat body temperature were analyzed. At the same time, the Kd and Bmax of D_1 dopamine receptor in rat striatum were identified with radioligand binding assay. The concentration of cAMP and the content of CaM were respectively examined by radio-competitive binding reaction and FACS. The results showed that Madopar accelerated the body temperature raise of rat suffering heat stress;the antagonist SCH23390, however, blocked the raise rate. The different effects of these drugs on thermoregulation may be induced by D_1,R and D_2, R and have different molecule mechanism.
