摘要
[目的]合成咪唑乙烟酸完全抗原并加以鉴定。[方法]以除草剂咪唑乙烟酸原药、草酰氯以及六氨基己酸等为起始原料,通过亲核取代、水解等反应合成了咪唑乙烟酸的半抗原。用碳二亚胺法将半抗原与牛血清白蛋白偶联制备免疫原,用混合酸酐法将半抗原与鸡卵清蛋白偶联制备包被原。[结果]半抗原的结构经IR、UV、1H NMR以及HPLC-MS确证。经紫外光谱分析,计算得出半抗原与牛血清白蛋白和鸡卵清蛋白的结合比为18∶1和7∶1。[结论]紫外光谱分析证实咪唑乙烟酸抗原合成成功,该抗原可用于咪唑乙烟酸多克隆抗体和ELISA检测试剂盒的研制。
[Aims] The study aims to synthesize and identify imazethapyr artificial antigen. [Methods] The hapten of imazethapyr was Synthesized using imazethapyr, oxalyl chloride and aminocaproic acid as original materials via nuclcophilic substitution and hydrolysis reaction. The hapten was conjugated with bovine serum albumin (BSA) by carbodimide method to obtain immunogen and with ovalbumin (OVA) via mixed anhydride method to get covering antigen. [Results] The structure of the hapten was confirmed by IR, UV, ^1H NMR and HPLC-MS. According to accumulation principle of UV absorption, the conjugated-ratios of BSA and OVA were 18 : l and 7 : 1. [Conclusions] The two artificial antigens were successfully synthesized, which provides a basis for next polyclonal antibody preparation and ELISA kit development.
