浮舰蛋白-1(Flotillin-1)是属于SPFH家族的蛋白,是重要的脂筏标志性蛋白。在构建大黄鱼(Larimichthyscrocea)肌肉组织cDNA文库的基础上,克隆了Flotillin-1基因,并进一步扩增出内含子。克隆到的序列全长为2497bp,其中编码区1194 bp,编码397个氨基酸。生物信息学分析大黄鱼Flotillin-1有5类20个功能位点,存在2次跨膜结构,N端和C端都位于细胞膜内。大黄鱼Flotillin-1氨基酸序列具有非常高的保守性,与大西洋鲑和斑马鱼的同源性都在80%以上。在组织中的表达也非常广泛,其中在脾中的表达最强。
Flotillin-1, one member of the SPFH protein family, is an important marker protein for lipid raft. On the basis of construc- ting the full length cDNA library from the skeletal muscle of Larimichthys crocea, flotillin-1 gene was cloned and sequenced. The intron sequences were also identified by PCR and sequencing. The acquired flotillin-1 gene was 2497 bp in length, and the coding sequence was 1194 bp in length, which encodes 397 amino acids. The bioinformaties analysis showed that flotillin-1 of Larimichthys crocea had twenty functional sites and had two hydrophobic transmembrane spanning domains. Both its N-and C-termini were located in the interior of cell membrane. The flotillin-1 amino acid sequence was highly conserved, and the similarities among Salmo salar, Danio rerio, and Larimichthys crocea were over 80%. The flotillin-1 gene was expressed in various tissues, with the highest expression in spleen, highly in brain, liver, eye, kidney and gill.
