摘要
用TTC法测定油茶花粉散粉后的活力变化及寿命,用琼脂培养基萌发法测定了花粉萌发率,用联苯胺-过氧化氢法测定了柱头可授性。结果表明:油茶花粉活力在散粉8 h呈上升趋势,即散粉第1~2天保持较高活力,随后活力开始下降,花粉寿命为4~10 d,最佳传粉期为开花后第1~5天;油茶花粉萌发率是体现花粉活力的一个指标,结果略低于TTC法测定的花粉活力值,它是对TTC法测定花粉活力的补充和验证;油茶柱头可授性持续3~6 d,花苞在将开而未开时已具备一定可授性,开花第1~5天柱头可授性呈上升趋势,第5~10天可授性减弱至基本消失;与柱头可授性相比,花粉寿命相对长些,试验结果与所观测的单花开放寿命较相一致。
TTC method was utilized to determine the vigor change and life-span of the Camellia oleifera pollen after anther dehiscence;with agar medium germination method,the pollen germination rate was determined;and with benzidine-hydrogen peroxide method,the stigma receptivity was measured.The results show that Camellia oleifera pollen vigor showed a rising trend in 8 hours after anther dehiscence,within 1~2 days the pollen maintained at a high vigor level,then gradually declined,the life-span of Camellia oleifer pollen was around 4~10 days or so,and the best pollination period was 1 to 5 days after flowering;The rate of Camellia oleifer pollen germination is an indicator of pollen vigor,the determining value was slightly lower than the result of the TTC method,which is a kind of supplement and verification to TTC method in determining the pollen vigor;The stigma receptivity maintained for 3~6 days or so,but the best pollination period was about 5 days,among open in the bud and the un-blossomed the flowers already have a certain receptivity,flowering the first 1~5 days the stigma receptivity was rising,5~10 days started to reduce until to basically disappeared;Compared with the period of stigma receptivity to pollen,life-span was relatively longer,the test results and the observation of a single flower opening life was consistent.
