摘要
目的通过对大量显微注射实践工作中相关数据的分析,寻求DNA片段长度与制备转基因小鼠整合效率的相关性的规律。方法随机抽样29个制备转基因的项目的资料进行相关性分析。结果制备转基因小鼠的整合效率随着DNA片段长度增加而降低。根据回归方程,如果片段长度能控制在6000bp以内基本上可以维持10%左右的整合阳性率(P〈0.001)。结论制备转基因小鼠的DNA片段长度与整合效率之间存在负相关。
Objective To analysis the relevance of foreign DNA length to integration efficiency on the randomly gathering data in preparation of transgenic mouse. Methods The statistics analysis was performed on 29 randomly gathered data of generation of transgenic mouse. Result According the regression equation of statistics analysis in this paper, the integration efficiency of foreign DNA was increased with the shorter length of foreign DNA (P〈0.001). The foreign DNA fragments integrated into mouse chromosome DNA with 10% efficiency when the length of foreign DNA were less than 6 000 bp. Conclusion There is a negative correlationbetween integration efficiency and foreign DNA length in microinjection.
