摘要
报道了灵敏度高、选择性好、简便快速的硝苯地平血药浓度的HPLC测定方法。采用Luna5μC18(2)柱,乙腈-水-二乙胺-磷酸(51∶49∶0.12∶0.10)为流动相,安定为内标,UV检测波长238nm,血浆最低检测浓度为2ng/ml,线性范围2~180ng/ml。
An improved HPLC determination of nifedipine in human plasma is reported. The chromatographic system consisted of Luna C18( 2 ) column and mobile phase of acetonitrile- water- diethyl- amine- H3PO4( 5 1∶ 4 9∶ 0 .1 2∶ 0 .1 0 ) . The detective wavelength was 2 38nm with diazepam as the internal standard. The plasma samples were extracted with ether and n- hexane ( 3∶ 1 v/ v) ( recovery>95 % ) . The detective limit was 2 ng/ ml,and the calibration curve was linear from 2 to 1 80 ng/ ml with a regression coefficientof0 .9996.
