摘要 目的探讨多药耐药基因MDR1在胃癌组织中的表达及临床意义。方法收集2009年12月~2010年8月胃癌新鲜癌组织标本53例(实验组)、15例正常胃黏膜组织(对照组),采用逆转录一聚合酶链反应技术检测胃癌组织和正常胃组织中MDR1基因的表达,并分析与临床病理特征之间的关系及对化疗的指导意义。结果MDR1基因在正常胃组织及胃癌组织中的表达分别为0.57±0.27和0.20±0.10(t=8.13,P〈0.001)。MDR1基因的表达与肿瘤分化程度、淋巴结转移之间差异具有统计学意义(t=2.83,3.47;P=0.007,0.002),而与患者性别、肿瘤部位、大小之间差异无统计学显著性意义(t=1.41,0.54,1.16;P=0.16,0.60,0.25)。结论MDR1基因的高表达可能是胃癌多药耐药产生的基础,MDR1基因的检测对制定化疗方案和选择化疗药物有一定的指导意义。 Objective To investigate the expression levels of MDR1 mRNA in patients with gastric cancer and clinical significance. Methods Collected in December 2009 to August 2010 fresh gastric cancer 53 cases(exprement group), 15 cases of normal gastric mucosa tissue(control group). RT-PCR method was used to detect the expression levels of MDR1 mRNA in gastric cancer cases and normal tissues,analyzed the relationship between the expression of MDR1 and the clinicopathologic paramenters. Results The expression level of MDR1 mRNA (0. 57±0. 27) was significantly higher in gastric cancer tissues than that in normal tissues (0. 20±0. 10) (P〈0. 001). The expression of MDR1 intensity was related to differentiation degree and lymph node metastasis ( t= 2.83,3.47 ; P= 0. 007,0. 002), had nothing to do with other clinicopathologic parameters( t=1.41,0. 54,1.16; P=0. 16,0. 60,0. 25). Conclusion The over-expression levels of MDR1 mRNA may underline the multidrug resistance of gastric cancer, the detection of expression can be looked as an importance method for judging the prognosis and guiding chemotherapy in gastric cancer.
