摘要
目的建立兔肢体缺血模型,观察二次外周血干细胞移植对肢体缺血的治疗效果。方法连续4 d皮下注射7.5μg/kgG-CSF进行干细胞动员,分离外周血单个核细胞制成干细胞悬液。制备兔肢体缺血模型,将干细胞悬液分10~12个点注射于手术切口两侧肌肉,实验组动物于移植后第18天重复进行干细胞动员并在第22天进行第二次干细胞移植。结果术后多普勒超声显示手术肢体的血液供应阻断,模型制备成功。1周后检测侧支循环开始建立,实验6周后,二次移植兔缺血部位肌肉毛细血管密度平均为26.1个,单次移植兔为20.9个,差异有统计学意义(P<0.05)。结论二次外周血干细胞移植能更好地改善肢体缺血。
Objective To construct the rabbits′model of limb ischemia and observe the effect of two-times autologous peripheral blood stem cell transplantation.Methods After injecting 7.5 μg/kg G-CSF every day for 4 days,then peripheral blood mononuclear cells and prepared stem cells suspension were separated.When the rabbits′model of limb ischemia was constructed,the stem cell suspension were injected into both sides of the surgical incision by 10 to 12 points.After the 18th days the stem cell mobilization was carried out again and was performed at the 22th day.Results The blood supply of surgery limb was blocked showed by doppler ultrasound,so the model was constructed successfully.After 6 weeks the ischemic muscle capillary density of two-times transplantation experimental rabbits was 26.1,and the control group was 20.9.The difference was statistically significant(P 0.05).Conclusion The two-times autologous peripheral blood stem cells transplantation can promote angiogenesis and improve limb ischemia.
