摘要
目的研究急性高眼压后大鼠视网膜血红素氧合酶-1(HO-1)表达的变化规律,为视网膜损伤后的治疗提供理论依据。方法采用前房灌注加压法制作急性高眼压大鼠模型,运用实时定量逆转录聚合酶链反应方法测定HO-1mRNA的表达情况。结果急性高眼压后1d视网膜HO-1mRNA的表达明显升高且达高峰,3d时表达水平下降,7d时再次升高,14d时继续升高。结论急性高眼压造成视网膜氧化应激损伤可以诱导HO-1表达增高。
Objective To study the law of changes of expression of HO-1 in rat retina following acute ocular hypertension(OH) and provide a theoretical evidence for the therapy of retinal injury. Methods An acute OH model in rats was created by perfusing normal saline into anterior chambers of rat eyes.The expression of HO-1 in rat retina was detected by using real-time reverse transcription polymerase chain reaction. Results The expression of HO-1 mRNA increased markedly one day after elevation of intraocular pressure and reached the peak,the expression then descended on the third day.On day 7,the expression increased again and until day 14. Conclusion Oxidative stress injury of retina due to acute OH may induce the elevation of HO-1 expression.
