摘要 目的:研究中药紫龙金逆转人膀胱癌细胞多药耐药(Multidrug resistance,MDR)机理。方法:应用MTT法检测紫龙金的细胞杀伤作用,采用流式细胞仪检测细胞凋亡,RT-PCR法检测Bcl-2、Bax基因mRNA水平表达。结果:BIU-87/ADM细胞生存率为(98.00±1.48)%,加入紫龙金后(浓度分别为200、500μg/ml),其细胞生存率分别为(68.14±5.62)%、(52.36±1.87)%,差异显著(P<0.01);流式细胞仪检测发现:经200、500μg/ml的紫龙金处理后,BIU-87/ADM细胞凋亡率[(17.32±2.35)%、(22.76±3.65)%]明显高于对照组(5.86±1.37)%,差异显著(P<0.01);RT-PCR结果显示,紫龙金可以下调Bcl-2基因的表达,可使Bax基因表达增强。结论:紫龙金可通过下调Bcl-2基因的表达,增强Bax基因表达诱导人膀胱癌多药耐药细胞凋亡。 Objective:To study the reversal of multi-drug resistance by Zilongjin in human bladder cancer cell line.Methods:The cytotoxicity of Zilongjin on human bladder cancer cell line BIU-87/ADM was studied by MTT assay.Apoptosis was detected by flow cytometry;expression of Bcl-2/Bax gene was studied by RT-PCR.Results:BIU-87/ADM cell survival rate was(98.00±1.48)%.After being treated with Zilongjin,cell survival rate was(68.14±5.62)% and(52.36±1.87)%,respectively(P0.01);apoptosis rate in cell line BIU-87 /ADM(treated with Zilongjin) was(17.32±2.35)% and(22.76±3.65)%,higher than that in control group cell line [(5.86±1.37)%,P0.01].Bcl-2 gene expression was decreased,while Bax gene expression was increased according to RT-PCR assay.Conclusion:Apoptosis can be induced by Zilongjin through decreasing Bcl-2 and increasing Bax gene expressions in human bladder cancer multi-drug resistance cell line.
