摘要 目的:研究马齿苋总黄酮的提取工艺。方法:采用正交设计法及聚酰胺吸附分离法,以提取物中总黄酮含量为指标,考察马齿苋的提取工艺;并应用分光光度法测定提取物中总黄酮含量。结果:马齿苋的最佳提取工艺为每次加40倍的70%乙醇溶液,提取2次,每次提取60 min,所得提取液中的总黄酮含量最高;并应用聚酰胺吸附分离其中的总黄酮,以芦丁为标准品,以510 nm为检测波长,用UV法检测马齿苋总黄酮在0.007-0.0764 mg/ml范围内,浓度与吸光度呈良好的线性关系,相关系数R2=0.9991,该法所得提取物中总黄酮的相对含量最高为6.936%。结论:应用醇提、聚酰胺吸附分离法提取马齿苋中的总黄酮,方法简便、准确,为鉴定马齿苋的质量提供了一种可行的方法。 Objective: To study the optimum extraction technique of total flavonoids in Purslane.Methods: To select the optimum extraction technique for Purslane,Total flavonoids extracted with orthogonal design and by polycaprolactam adsorption method.Total flavonoids in Purslane were determined using UV spectrophotometry.Results: The optimum process of alcohol-decoction was to add 40 times amount of 70% alcohol,decocting for 2 times,60 minutes each time.the lever of total flavo-noids in hyperin was the highest.The contrast sample was rutin for ployamide-UV spectrophotometry.The optimum determination wavelength was 510nm.the result showed that a good linearity relationship(correlation coefficient:0.9991)between the degree of absorbance and the concentration of analyte was obtained,within the range of 0.007-0.0764mg/ml.the lever of total flavonoids in extracting agent was the highest using polycaprolactam,It's 6.936%.Conclusion: To extract flavonoids from Purslane by using alcohol-dococting and polycaprolactam,the method is convenient and exact.
