摘要
目的通过检测α-actinin-4mRNA和nephrinmRNA在糖尿病肾病(DN)患者肾小球组织中的表达,探讨其在糖尿病肾病患者蛋白尿发生中的作用。方法用实时荧光定量逆转录聚合酶链反应(RT—PCR)技术检测α-actinin-4mRNA和nephrinmRNA在测糖尿病肾病组、单纯糖尿病组、单纯血尿组及对照组患者肾小球组织中的表达。结果糖尿病肾病组nephrinmRNA表达显著低于单纯糖尿病、单纯血尿组及对照组,糖尿病肾病组α-actinin-4mRNA表达显著低于单纯糖尿病、单纯血尿组及对照组,两者表达存在正相关。结论糖尿病肾病患者肾小球组织nephrin和α-ctinin-4mRNA的表达减少,导致蛋白尿的产生。
Objective To detect the expression of a-actinin4 mRNA and nephrin mRNA in dia- betic nephropathy(DN), and to investigate their significance in the development of proteinuria in DN. Methods Real-time fluorescent quantitative RT-PCR was used to detect α-actinin4 mRNA and nephrin mRNA of glomerulus from diabetes nephropathy ( DN), diabetes mellitus ( DM ), isolated hematuria and healthy control group. Results The expression of nephrin mRNA in the glomerulus decreased significant- ly comparing with that in DM patients ( P 〈 0.05 ), isolated hematuria patients ( P 〈 0.05 ), and control group (P 〈 0.05 ). And the same result was received from the expression of α-actinin-4 mRNA in the glo- merulus. The Spearman rank correlation analysis demonstrated a significant correlation between α-actinin- 4 and nephrin expression(r = - 0. 405,P 〈 0.05). Conclusion Both the dramatic decreasion of the ex- pression of nephrin mRNA and α-actinin-4 mRNA might contribute to the development of proteinuria from patients with DN.
