摘要
目的研究3型鼠肝炎病毒诱导的小鼠病毒性肝炎模型中肾脏组织肿瘤坏死因子-α的表达及其意义。方法建立MHV-3病毒诱导的小鼠病毒性肝炎模型,测定血清尿素氮、肌酐水平;采用免疫组化方法检测肾脏组织肿瘤坏死因子-α的表达;逆转录-聚合酶链反应法检测肾脏组织肿瘤坏死因子-α的mRNA水平。结果 MHV-3病毒感染小鼠后各时间点血清BUN、Cr水平均较正常对照明显著升高。模型小鼠肾脏组织肿瘤坏死因子-α的表达,随着MHV-3病毒感染时间的延长,肾脏组织中肿瘤坏死因子-αmRNA水平逐渐增高,在第10天达到高峰。结论肾脏组织表达的肿瘤坏死因子-α可能参与MHV-3病毒感染引起的肾脏损伤的病理过程。
Objective To investigate the expression and significance of TNF-α in the kidney of MHV-3 induced murineviral hepatitis model. Methods The levels of urea nitrogen (BUN) and serum creatinine (Cr) in serum were detected, andthe expression of TNF-α in kidney tissue were detected by immunohistochemistry and RT-PCR respectively. Results Bythe time-course of MHV-3 infection, levels of BUN and Cr in serum were elevated significantly. TNF-α was detectable inkidney tissue after MHV-3 infection. The level of TNF-α mRNA in kidney were elevated significantly, and reached highestlevel 10 days post MHV-3 infection. Conclusion The increased expression of TNF-α may play an important role in thepathogenesis of renal injury induced by hepatitis viral infection.
