热应激对猪小肠组织形态和细胞凋亡的影响 被引量:14

The research of injury and apoptosis in the swine small intestine epithelium treated by heat stress

摘要 采用HE染色观察猪小肠形态学变化,经PCR对猪小肠细胞凋亡基因Caspase-3、8、9、Bcl2、Bax的变化进行研究,免疫组化检测Caspase-3在小肠中的表达情况,探明热应激致小肠组织损伤的分子机制。结果:在热应激中,猪的小肠绒毛顶端上皮细胞脱落;Caspase-3、8、9、Bax基因表达上调,Bcl2基因表达下调;Caspase-3蛋白在小肠绒毛顶端表达升高。结论:热应激过程中,猪的小肠形态学损伤,细胞凋亡明显,绒毛顶端凋亡严重;死亡受体、线粒体途径被激活,促凋亡作用增强,抗凋亡作用减弱。 This research was to investigate the alteration of the morphological changes, the expression of apoptotic genes and proteins in the small intestine of swine, to provide the indexes of apoptosis treated by heat stress, and to provide the drug's functional targets and evaluative factors. Compared to the control, the tips of the villi shed into the lumen, the expression of the genes of Caspase--3, Caspases--8, Caspase--9 and Bax in the heat stress group improved , Bcl2 expressed lower, the pro- tein Caspase-3's expression advanced in the head of the intestinal villi. The result indicated that the completeness of the intes-tine was destroyed, the level of apoptosis in the cells of the small intestine got higher, especially in head of the intestinal villi, and the extrinsic and intrinsic pathway were activated.
