草鱼肝脏细胞色素P450酶系生物转化酪胺生成章鱼胺的研究

Biotransformation from tyramine to octopamine catalyzed by cytochrome P450 in hepatic S-9 position of grass carp

摘要 目的以草鱼肝脏S-9部位为酶源,以酪胺为原料生物转化生成章鱼胺。方法制备草鱼肝的S-9部位,测定细胞色素P450酶的含量并作生物转化试验。以维生素C为氢载体,异烟肼为单胺氧化酶抑制剂。结果反应1h产生的章鱼胺为879.7μg/4.5mL反应液,章鱼胺的生物合成率为21.4%,酪胺代谢率为90.0%。结论以草鱼肝脏S-9部位为酶源生物转化生成章鱼胺是可行的,但由于P450酶系的非专一性,章鱼胺的生物合成率较低,有待进一步改进。 Objective To generate octopamine, using tyramine as substrate and grass carp hepatic S-9 position as the resource o f cytochrome P450. Methods After hepatic 8-9 position of grass carp had been prepared and the content of cytochrome P450 had been determined, tyramine was biotransformed to octopamine with ascorbic acid as hydrogen carrier and isoniazid as monoamine oxidase inhibitor. Results The octopamine obtained after 1 h catalysis was 879.7μg in 4. 5 mL reaction liquid. The biosynthesic rate of octopamine and metabolic rate of tyramine were 21.4% and 90. 0%, respectively. Conclusion It is feasible to produce octopamine when hepatic S-9 position of grass carp being taken as resource of enzyme. But because of the non-specifity of cytochrome P450 the biosynthesic rate of octopamine was relative low, which need to be further improved.
