薯蓣皂苷对离体大鼠主动脉环的作用研究 被引量:1

摘要 目的:观察薯蓣皂苷对离体大鼠主动脉环的舒张作用。方法:采用离体器官水浴法,分为3组,分别为对照组、完整内皮组和去内皮组,观察薯蓣皂苷(0.1~3.2 mmol/L)对苯肾上腺素(PE)、氯化钾(KCl)预收缩的大鼠主动脉环的作用,进行组间比较,并观察薯蓣皂苷对离体血管作用的内皮依赖性。结果:(1)对PE预收缩内皮完整血管环,薯蓣皂苷在0.1 mmol/L即产生显著舒张作用(vs对照组P〈0.01)且有剂量依赖性;(2)对PE预收缩去内皮血管环,薯蓣皂苷在1.6 mmol/L产生舒张作用但效果不显著(P〉0.05),在3.2 mmol/L产生舒张作用(P〈0.05);(3)在相同浓度薯蓣皂苷(0.1~0.8 mmol/L)条件下,PE对内皮完整组舒张程度显著高于去内皮组(P〈0.05),表明薯蓣皂苷对PE预收缩的血管环舒张作用有一定的内皮依赖性;(4)对KC1预收缩内皮完整血管环,薯蓣皂苷在0.8 mmol/L产生显著舒张作用(vs对照组P〈0.01)且有剂量依赖性;(5)在KC1预收缩的去内皮血管环,薯蓣皂苷1.6 mmol/L产生舒张作用但效果不显著(P〉0.05),薯蓣皂苷3.2 mmol/L产生舒张作用(P〈0.05);(6)在相同浓度薯蓣皂苷(0.8~1.6 mmol/L)条件下,薯蓣皂苷对内皮完整主动脉环的舒张作用显著高于去内皮组(P〈0.05),表明薯蓣皂苷对KC1预收缩的血管环舒张作用有一定的内皮依赖性。结论:薯蓣皂苷通过内皮和非内皮作用机制舒张血管,并具有剂量依赖性。 Objective: To observe the effects of dioscin on aortic vascular rings isolated from rat.Methods: The aortic vascular rings were perfused with water bath method and divided into 3 groups,namely as control group,complete endothelial group and none endothelial group.The effects of dioscin(0.1~3.2 mmol/L) on the vascular ring pre-contracted with phenylephrine(PE),or potassium chloride(KCl) were observed,under the condition of endothelium intact and without endothelium.Results:(1)Under the conditions of vascular endothelial integrity,dioscin produced a significant vasodilation from 0.1 mmol/L(vs control group P0.01) on the vascular rings pre-contraction with PE,and possessed a dose-dependent vasodilation function;(2) Under the conditions of none vascular endothelial,dioscin produced diastolic function but not significant in 1.6 mmol/L(P0.05) on the vascular rings pre-contraction with PE,and produced vasodilation function in 3.2 mmol/L(P0.05);(3) Under the conditions of the same concentration of dioscin(0.1~0.8 mmol/L),the degree of vasodilation was significantly higher in the complete vascular endothelial group than that in the none vascular endothelial one(P0.05) on the vascular rings pre-contraction with PE,which shown that the dioscin possessed a endothelial – dependent vasodilation function on PE pre-contraction of vascular rings;(4) Under the conditions of vascular endothelial integrity,dioscin produced a significant vasodilation from 0.8 mmol/L(vs control group P0.01) on the vascular rings pre-contraction with KCl,and posessed a dose-dependent diastolic function;(5) Under the conditions of none vascular endothelial,dioscin produced diastolic function but not significant in 1.6 mmol/L(P0.05) on the vascular rings pre-contraction with KCl,and produced diastolic function in 3.2 mmol/L(P0.05);(6) Under the conditions of the same concentration of dioscin(0.8-1.6 mmol/L),the degree of vasodilation was significantly higher in the complete v
