摘要
目的研究视觉发育敏感期内,剥夺性弱视大鼠视觉系统神经生长因子的表达规律,探讨其在弱视发病机制中的作用。方法缝合大鼠单侧眼睑30d,建立剥夺性弱视模型。应用HE染色法观察外侧膝状体神经元的形态改变;免疫组化技术检测神经生长因子在大鼠外侧膝状体的表达情况。结果免疫组化结果显示:剥夺眼对侧外侧膝状体组较剥夺眼同侧外侧膝状体组及正常对照组神经生长因子的表达明显减少,差异有显著性(P<0.05)。结论视觉发育敏感期内,形觉剥夺后外侧膝状体神经生长因子的表达降低,可能引起了神经元的分化与成熟障碍,导致神经元之间的突触连接减少可能是弱视发生的机制之一。
Objective To investigate the regulation of expression of nerve growth factor(NGF)at lateral geniculate body(LGB)of monocular form-deprivation amblyopia rats within sensitve period of visual development,in order to explore its role in the pathogenesis of amblyopia.Methods We sutured unilateral eyelids on experimental group rats for 30 days and the models of form-deprivation amblyopia were established.We observe the neural morphological change of the LGB in HE staining,and detect the expression of NGF at LGB of rats in immunohistochemistry technology.Results Of immunohistochemistry: Compared with normal control group and ipsilateral LGB of deprived eyes group,the expression of NGF at contralateral LGB of deprived eyes group are remarkablly decreased,and the differences were significant(P0.05).Conclusions In the sensitve period of visual development,the expression of NGF are depressed after form-deprivation,which may induce dysdifferentiation and dysmaturity of neurons,then the synaptic linkage decreased and this may be one of the pathogenesis of amblyopia.
