摘要
本研究确定的体外透析管法测定饲料蛋白质和氨基酸消失率的适宜酶促反应条件是 :温度 3 5℃ ,胃蛋白酶溶液浓度 0 .0 0 1 g/ml,胰蛋白酶溶液浓度 0 .0 0 2 5g/ml( p H7.6) ,透析液体积 3 0 0 ml( 1 .0 0 0 0 g样品 ) ,胰蛋白酶处理时间2 4 h。依据体外透析管法的测定结果 ,建立了估测饲料蛋白质和氨基酸消化率的回归预测模型。
In this paper, a new in vitro method of determining the digestibilities of crude protein and amino acids using the dialysis tube for pigs under the laboratory conditions has been set up. The procedures of the in vitro dialysis tube method are as flooows: weighing accurately 1 0000g sample and transferring them into a flask, then adding pepsin solutions (the pepsin concentration is 0.001g/ml) and hydrolysising for 4 hours, then neutralizing the solution with caustic soda, and transferring them into a dialysis tube, and adding trypsin solutions (pH is equal to 7.6 and the trypsin concentration is 0.0025g/ml) and hydrolysising for 24 hour at 35 ℃. The in vitro and in vivo experimental results show that it occurred significantly positively correlative between the in vitro disappearance and the in vivo digestibility of crude and amino acids for pigs, so it is feasible to predicted the in vivo digestbilities of crude protein and amino acids for pigs.
