摘要
目的建立测定人血浆中头孢克肟含量的离子对反相高效液相色谱法。方法血浆样品用10%三氯醋酸沉淀蛋白,以替硝唑为内标,经Capcell Pak UG120 C18色谱柱分离,流动相为乙腈:0.02 mol.L-1庚烷磺酸钠(18∶82,pH 3.0),流速1.0 mL.min-1,检测波长290 nm。结果头孢克肟浓度在0.05~5.00 mg.L-1内线性关系良好(r=0.996 8),日内、日间RSD均≤15.0%,提取回收率在54.76%~59.85%之间。结论本方法准确、快速,适用于头孢克肟的血药浓度测定及药代动力学研究。
[ Objective] A reversed phase ion pair HPLC method was established for determination of cefixime in human plasma. [ Methods] Plasma was precipitated with trichloroacetie acid, using tinidazole as internal standard. The analysis was conducted with a Capcell Pak UG120 C18 column. The mobile phase consisted of acetonitrile and 0. 02 mol · L^-1 sodium heptanesulfonate ( pH 3.0, 18:82 ). The flow rate was 1 mL ·min^-1 , and the detecting wavelength was at 290 nm. [ Results] The linear range cefixime was 0.05 -5.0 mg · L^-1 and the method recoveries were within 54.76 % - 59.85 % at three different concentrations. The RSD of intra - and inter - day were all less than 15 %. [ Conclusion] This method is simple , accurate and reliable . It is suitable for the pharmaeokinetic study of cefixime.
