侵染云南烟草的番茄环纹斑点病毒N基因的分子变异分析 被引量:12

Molecular variation of natural populations of Tomato zonate spot virus infecting flue-cured tobacco in Yunnan

摘要 应用电镜观察和ELISA检测从云南11个县(市)烟草种植区采集的烟草上检测到番茄环纹斑点病毒(Tomato zonatespot virus,TZSV)。根据TZSV N基因设计引物扩增得到了31个TZSV分离物N基因全序列。测序分析表明,31个TZSVN基因核苷酸序列与GenBank中报道的序列相似性在94.9%~98%之间,其推导的氨基酸序列同源性为98.2%~99.3%。构建系统进化树发现云南不同地区的TZSV分离物存在一定差异。氨基酸序列比较发现,文山分离物和西畴分离物具有2个该地区独有的氨基酸位点。 Using electron microscopy and ELISA, Tomato zonate spot virus (TZSV) isolates were detected from flue-cured tobacco in 11 counties and cities of Yunnan. With primers from conserved regions, the nucle-ocapsid (N) gene sequences of 31 naturally occurring TZSV isolates from flue-cured tobacco were amplified by RT-PCR. Sequence analysis showed a high level of sequence conservation (94.9% -98% ) among the N genes of the tobacco isolates. However, cluster dendrogram showed distinct differences were present in Yun-nan TZSV isolates from different area. Amino acid sequence analysis showed that two amino acid residues were considered to be area specific in Wenshan isolates and Xichou isolates.
