摘要
用高效液相色谱法测定分蘖葱头总黄酮微乳中的槲皮素含量。采用Shmisadezu LC-10ATvp高效液相色谱仪,YMC-PackProC18RS(I.D.250mm×4.6mm)色谱柱,流动相:甲醇-0.4%磷酸溶液(51:49);检测波长:365nm;流速:1.0mL/min。结果表明,槲皮素在0.268~85.816μg/mL范围内线性关系良好,平均回收率98.03%,RSD1.02%。该方法简单、快速、准确、重复性好,可用于测定分蘖葱头总黄酮微乳中的槲皮素含量。
The aim of this study was to determine the contents of quercetin in microemulsion of flavonoid-rich allium cepa L. var. agrogatnm don extract. High performance liquid chromatograph (HPLC) was used in this experiment. The condition as follows: Shmisadezu LC-10A Tvp (250mm×4.6mm) was adopted, mobile phase consisted of methanolwater (51:49), the wavelength was 365nm, and the flow rate was l.OmL/min. The calibration curve of quercetin was in good linearity with the range of 0.268-85.816 μ g (r=0.999 9). The average recovery was 98.03%, and RSD was 1.02%. The HPLC method is simple, stable, accurate and reproducible, and it can be used for determining the content of quercetin in microemulsion of flavonoidrich allium cepa L.var. agrogatnm don extract.
