染尘大鼠模型中炎症递质的变化和意义 被引量:2

CHANGES AND SIGNIFICANCE OF INFLAMMATORY TRANSMITTERS IN DUST-EXPOSED RAT MODEL

摘要 目的观察矽肺形成过程中炎症递质肿瘤坏死因子-α(TNF-α)、白细胞介素-8(IL-8)和高敏C-反应蛋白(hsCRP)的动态变化和意义,探讨炎症因素在矽肺发病过程中的作用。方法选取Wistar大鼠64只,随机分为模型组、对照组,每组32只。模型组气管滴注二氧化硅混悬液建立染尘大鼠模型,分别在染尘后的第3、7、14、28天分批处死大鼠,切取肺组织标本作病理切片,行苏木精-伊红(HE)染色、MASSON染色,观察肺组织病理变化。用酶联免疫吸附试验(ELISA)法测定血清和支气管肺泡灌洗液(BALF)中TNF-α和IL-8的含量,用散射比浊法测定hsCRP含量。结果模型组大鼠血清和BALF中TNF-α、IL-8、hsCRP的浓度均明显高于对照组,差异有显著性(t=2.64~22.26,P<0.05)。BALF和血清中的IL-8、hsCRP浓度高峰位于早期(第3天或第7天),BALF中TNF-α浓度高峰位于第14天(F=24.89~263.37,q=5.07~27.51,P<0.01)。结论在染尘大鼠模型中,IL-8、TNF-α和hsCRP是气道和全身主要的炎性递质,在矽肺纤维化早期发挥着重要作用。 Objective To investigate the dynamic changes and significance of tumor necrosis factor(TNF-α),interleukin 8(IL-8) and hsCRP in formation of silicosis,and study the role of inflammation in the development of this condition.MethodsThis study consisted of 64 Wistar rats,which were evenly randomized to model and control group.A dust-exposed rat model was created by intratracheal drip of silicon dioxide suspension,which were killed at days 3,7,14,and 28 upon completion of the model.The lung tissues were collected for pathology.HE and Masson staining were applied.The levels of TNF-α and IL-8 in serum and bronchovesicular lavage fluid(BVLF) were detected by using ELISA and the levels of hsCRP by nephelometry.Results The levels of TNF-α,IL-8 and hsCRP in BVLF and serum in the model group were higher than that in the control group(t=2.64-22.26,P〈0.05).The concentration peak of IL-8 and hsCRP in BVLF and serum was in early phase(day 3 or day 7),and that of TNF-α in BVLF was on 14th day(F=24.89-263.37,q=5.07-27.51,P〈0.01).Conclusion In the dust-exposed rat model,TNF-α,IL-8 and hsCRP were the major inflammatory transmitters,which played an important role in the early stage of fibrosis of silicosis.
