摘要
目的:本论文主要研究抗癌药物卡铂在体外与长度为1341bp的DNA链相互作用的机理。运用原子力显微镜来直接观察卡铂和DNA分子作用后引发的DNA的结构、形貌和长度的变化,来阐明卡铂和DNA的作用方式。方法:卡铂和DNA分子首先在溶液中相互作用,然后取少量作用后的溶液用于原子力显微镜的观察。结果:卡铂与DNA相互作用后,DNA链发生弯曲,长度缩短,且随着浓度提高,长度缩短程度变大。
Objective:In present study,interactions of carboplatin and a linear DNA of 1341bp have been investigated by Atomic force microscopy(AFM). The changes of DNA structures and the length induced by different concentration of earbopl atin have been investigated. Methods: Carboplatin solution and DNA molecules were mixed, and incubated for different time. The obtained solution was used for preparing AFM sample. Results:The results indicated that both contour length and persistence length of DNA are shortened after interaction with earboplatin.
