丹参联合川芎嗪对肺纤维化大鼠肺组织JAK1、STAT1、ICAM-1表达的影响 被引量:17

EFFECTS OF COMBINATION OF SALVIA AND LIGUSTRAZINE ON EXPRESSION OF JAK1,STAT1,AND ICAM-1 IN LUNG TISSUE OF RAT WITH PULMONARY FIBROSIS

摘要 目的:观察丹参联合川芎嗪腹腔注射对博来霉素(bleomycin,BLM)致肺纤维化大鼠肺组织蛋白酪氨酸激酶1(JAK1)、信号转导与转录活化因子1(STAT1)、细胞间粘附分子-1(ICAM-1)表达的影响。方法:将90只成年雄性SD大鼠随机分为以下6组:生理盐水(NS)组、BLM组、地塞米松(DM)组、小剂量中药(A)组、中剂量中药(B)组、大剂量中药(C)组,每组大鼠各15只。BLM组、DM组、A组、B组、C组气管内灌注BLM复制大鼠肺纤维化模型,NS组气管内灌注等体积的NS,造模成功24h后各组大鼠给予药物腹腔注射。NS组和BLM组每日给予腹腔注射NS,DM组每日给予腹腔注射DM,A组、B组及C组每日给予不同剂量丹参和川芎嗪复合制剂腹腔注射,分别于造模后第7天,第14天和第28天处死每组大鼠各5只,肺组织多聚甲醛固定后行HE染色和Masson染色观察肺泡炎及肺纤维化程度;免疫组织化学法测定肺组织中JAK1、STAT1、ICAM-1的表达;血清用于肝肾功能的检测以评价药物安全性。结果:①大鼠血清肝肾功能检测结果显示各组两两比较差异无统计学意义(P>0.05)。②NS组大鼠各时间点肺组织未发现肺泡间隔水肿、炎性细胞浸润及纤维化的形成。BLM组大鼠第7天时肺泡腔内可见大量炎性细胞浸润,第14天时炎性细胞减少,肺泡间隔内成纤维细胞显著增多,肺泡结构破坏,第28天时肺纤维化程度进一步加重,形成广泛纤维化。与BLM组比较,DM组大鼠各时间点肺泡炎及纤维化程度减轻(P<0.05)。A组大鼠肺泡炎和肺纤维化程度与BLM组类似。B组、C组第14天肺泡炎及肺纤维化程度明显低于BLM组(P<0.05)。B组、C组第28天肺纤维化程度明显低于BLM组(P<0.05)。③与NS组比较,BLM组、DM组、A组、B组、C组肺组织中JAK1、STAT1、ICAM-1的表达水平明显升高(P<0.05)。④与BLM组比较,DM组及B组、C组肺组织中JAK1、STAT1、ICAM-1的表达水平明显降低(P<0.05)。⑤肺组织JAK1、STAT1、ICA Objective: To investigate the effects of Salvia combined with Ligustrazme on expression oI janus kinase I(JAK1), signal transducers and activatiors of transcription I(STAT1) and intercellular adhesion molecule- 1 (ICAM-1) in lung tissue of rat with pulmonary fibrosis induced by bleomycin (BLM). Methods: 90 adult male SD rats were randomly divided into six groups: normal saline (NS) group, BLM group, dexamethasone (DM) group, small dose of Chinese crude drug (A) group, medium dose of Chinese crude drug (B) group, large dose of Chinese crude drug (C) group, and BLM group. DM group, group A, group B and group C were intratracheally instilledwith BLM, while NS group was with NS. After 24 hours NS group and BLM group were intraperitoneally injected with NS, DM group was with dexamethasone, and group A, group B and group C were with different doses of Salvia Ligustrazine mixture. Five rats of each group were killed respectively at the 7th.14th and 28th day after intratracheal instillation. Lung tissue was fixed by paraform aldehyde, and HE and Masson staining wereperformed to investigate alveolitis and fibrosis; JAK1, STAT1 and ICAM-1 in lung tissue were assessed by immunohistochemistry; Serum was used to detect the liver and kidney function. Results: ①The value of the liver and kidney function in each group had no significant difference (P〉0.05). ②Alveolar septal edema, inflammatory cell infiltration and fibrosis were not found in NS group. In BLM group, lots of inflammatory cells infiltration wereobserved at the 7th day. Inflammatory cells were reduced, the number of the fibroblasts and matrix in alveolar septum were obviously increased, and alveolar structure was damaged at the 14th day. Pulmonary fibrosis was increased, severe fibrosis was found at the 28th day.
