摘要
目的:通过优化手段筛选脂质体最佳处方工艺,制备伊立替康脂质体,并对其进行质量评价。方法:采用薄膜分散法,改良薄膜分散法,逆向蒸发法,主动载药等方法制备伊立替康脂质体,通过包封率测定优选出最佳制备方法。采用正交试验设计,以包封率为考察指标,优化伊立替康脂质体的处方和工艺。结果:用最优处方制得的脂质体外观好,包封率达到90%以上,透射电镜观察外观圆整,均匀,粒径小于20nm。结论:改良薄膜分散法用于制备水溶性药物伊立替康脂质体是一种较好的制备方法。
Objective:To determine optimized prescription and technique for preparing Irinotecan liposomes,and then to evaluate its quality.Methods:The preparation methods of liposomes such as film dispersion,improved film dispersion,reverse phase evaporation,alcohol injection and ammonium sulfate transmembrane gradients were explored in order to select suitable technique.The orthogonal design was adopted to screen the formulation of Irinotecan liposomes.Results:The appearance prepared by optimized method was satisfactory.The entrapment efficiency is more than 90% and the particle size of Irinotecan liposomes was less than 20nm.Conclusions:Improved film dispersion method is an effective and feasible method for water-soluble drug Irinotecan.
