摘要
目的探讨淋巴细胞主动免疫治疗对原因不明性复发性流产患者Treg和Th17转录因子(Foxp3、ROR-γt)和功能因子(EBi3、IL-17A)的影响。方法确诊为原因不明性复发性流产的患者20例,并行一个疗程(3次)的淋巴细胞主动免疫治疗,采用逆转录聚合酶链反应(RT-PCR)法检测治疗前后患者外周血中Foxp3及ROR-γt因子mRNA的表达,酶联免疫吸附试验(ELISA)测定血清中白介素(IL)17A及IL-35的含量。结果治疗前后Foxp3 mRNA的相对表达量分别为(0.28±0.06)、(0.52±0.0);ROR-γt mRNA的相对表达量分别为(0.60±0.11)、(0.42±0.09);IL-17含量分别为(16.12±4.16、14.72±1.98)ng/L;IL-35含量分别为(11.53±2.26、26.34±2.77)ng/L,差异均有统计学意义(P<0.05)。结论治疗前后Treg和Th17细胞因子存在显著差异,且Treg因子变化更为明显,提示在妊娠结局中Treg细胞可能起较重要的作用。
Objective. To investigate the effect of lymphocyte immunotherapy on transcriptional (Foxp3, ROR- 7t) and functional (EBi3, IL-17A) cytγkines of Treg and Th17 cells in unexplained recurrent spontaneous abortion patients. Methods. Twenty patients suffered with unexplained recurrent spontaneous abortion were treated with active lymphocyte immunotherapy for 3 times. Reverse transcription-polymerase chain reaction (RT-PCR) was used to detect Foxp3 and ROR-~/t mRNA in peripheral blood. Enzyme-linked immunosorbent assay (ELISA) was used to measure IL-17 and IL-35 levels in serum. Results. The relative expressions of Foxp3 mRNA were 0. 28±0. 06 and 0.52±0.04 before and after the treatment, and 0.60±0.11 and 0.42±0.09 for ROR-Tt mRNA. While the serum levels of IL-17 were (16.12±4.16) ng/L and (14.72±1.98) ng/L, IL-35 levels as (11.53±2.26) ng/L and (26.34±2.77) ng/L. All the differences before and after treatment were statistically significant (P〈0.05). Conclusion. The levels of cytokines of Treg and Th17 cells were significantly different after treatment. The levels of Treg cytokines were changed more pronounced, it suggested that Treg cells might play more important role in the outcome of pregnancy.
