摘要
目的建立麻杏石甘汤高效液相色谱指纹图谱,采用多波长切换检测技术全面分析该复方指纹图谱,提高其质量控制。方法运用RP-HPLC多波长切换技术,采用C18色谱柱(250 mm×4.6 mm,5μm);流动相为乙腈-0.1%磷酸,进行梯度洗脱;检测波长为0~34 min:215nm;34~40 min:350 nm;40~50 min:254 nm;柱温:25℃,流速:1 ml/min。结果运用多波长切换技术所得指纹图谱方法学考察结果良好,采用色谱指纹图谱相似度评价软件,对麻杏石甘汤多波长切换波长指纹图谱进行相似度计算,其10批样品相似度在0.74~0.92之间,说明多批次样品具有代表性。结论多波长切换技术的运用实现了指纹图谱的全面和整体评价,能全面体现特征吸收波长相差较大的多个成分的指纹信息,为有效提高麻杏石甘汤质量控制提供参考。
Objective To set up a method for Maxing Shigan Decoction fingerprint detection by the Multi - Wavelength Switching technique in order to acquire more information about Maxing Shigan Decoction. Methods RP - HPLC was used to establish fingerprints with C18 column and Acetonitrile -0. 1% Phosphoric acid aqueous, a gradient eluting agent. Wavelength:0 - 34min 215nm;34 - 40min 350nm; 40 -50min: 254nm. Results Methodological study showed multi -wavelength switching technique could fully reflect the fingerprint characteristics which components has different absorption wavelength. Using chromatographic fingerprint evaluating software, calculate Maxing Shigan Decoction Multi -Wavelength Switching fingerprints similarity, the 10 batches of samples similarity were between 0.72 - 0.94. Conclusion Multi - wavelength switching enforces the overall evaluation of Maxing Shigan Decoction and provides a suggestion for enhancing the Maxing Shigan decoction quality control.
