摘要
目的:探讨补肾宣肺活血方对博莱霉素所致大鼠肺纤维化的干预作用及可能的机制。方法:SD大鼠经气管内灌注博莱霉素(5mg/kg)诱导肺纤维化制作动物模型,每天分别用补肾宣肺活血方灌胃(中药组)、氢化可的松琥珀酸钠腹腔注射(激素组,25mg/kg)、生理盐水进行干预(模型组、空白组)。空白组气管内灌注和灌胃均用生理盐水,4组动物均于药物干预后第7、14天分别处死,用苏木素-伊红(HE)染色观察肺组织病理学变化和免疫组化(SABC法)测定肺组织碱性成纤维细胞因子(bFGF)、肿瘤坏死因子-α(TNF-α)的表达。结果:中药组经治疗后肺泡炎症病变及肺纤维化程度明显轻于模型组,与激素组比较未见明显差异,但较空白组严重。中药组bFGF、TNF-α的表达亦低于模型组,但高于空白组。结论:补肾宣肺活血方可明显减轻博莱霉素诱导的肺纤维化的病变程度,其作用机制可能是通过降低bFGF、TNF-α的表达来实现的。
Objective:To study the effect and mechanism of Bushen Xuanfei Huoxue Recipe(BXH)for interfering bleomycin-induced pulmonary interstitial fibrosis in rats.Methods:Animal models of pulmonary fibrosis were established by intratracheal perfusion of bleomycin(5mg/kg) in SD rats.Subsequently,the model rats were given gastric gavage of BXH(BXH group),intraperitoneal injection of hydrocortisone sodium succinate 25mg/kg(hydrocortisone group)was given),and intratracheal and gastric perfusion of saline(blank group).The rats were executed for examination on day 7 and 14 of invention.Pathological changes of lung tissue were detected by hematoxylin-eosin(HE) staining,and the expressions of basic fibroblast growth factor(bFGF) and tumor necrosis factor-α(TNF-α)in lung tissue were determined by immunohistochemical technique.Results:The pathological changes of airsacculitis and fibrosis in BXH group were obviously slighter than those in the model group,while remarkably higher than that in the blank group.There were no obvious differences of airsacculitis and fibrosis between BXH group and hydrocortisone group.The protein expression of bFGF and TNF-α in BXH group was obviously lower than that in the model group,slightly higher than that in hydrocortisone group,while remarkably higher than that in the blank group.Conclusion:BXH can reduce the pathological changes of pulmonary fibrosis induced by bleomycin,and the therapeutic mechanism is probably related with the decrease of bFGF and TNF-α protein expression.
