浙产何首乌根与叶中大黄素含量比较 被引量:3

摘要 目的建立测定浙产何首乌根与叶中大黄素含量的高效液相色谱法,并比较根与叶中大黄素的含量。方法采用超声提取法制备供试品溶液,应用高效液相色谱法测定何首乌根与叶中大黄素的含量。色谱柱为Zorbax Ecilpse XDB-C18柱(150 mm×4.6 mm,5μm),流动相为甲醇-1%冰醋酸(70∶30),检测波长为254 nm,流速为1.5 mL/min,柱温为40℃,进样量为20μL。结果大黄素质量浓度在1~500μg/mL范围内与峰面积线性关系良好(r=0.999 5),平均加样回收率为99.71%,RSD为2.52%(n=6)。浙产何首乌根与叶中大黄素含量分别为2.094 7 mg/g和0.030 3 mg/g。结论该法适用于何首乌中大黄素含量的检测。浙产何首乌根与叶中大黄素含量差异显著。 Objective To determine and compare emodin in roots and leaves of Polygonum multiflorum Thunb. from Zhejiang province. Methods The sample solutions were prepared by ultrasonic extraction. HPLC method was used to determine emodin in roots and leaves of Polygonum multifiorum Thunb. Zorbax XDB-C18 column was used and the mobile phase was CH3- l%CH3COOH(70:30). The detection wavelength was 254 rim,the flow rate was 1.5 mL/min,the column temperature was 40℃ ,and the injection volume was 20 μL. Results Emodin showed a good linear relationship at the range of 1-500μg/mL( r =0. 999 5),and its average recovery rate was 99.71%, RSD was 2.52% (n=6).The contents of emodin in roots and leaves of Polygonum multiflorum Thunb. from Zhejiang province were 2. 094 7 mg/g and 0. 030 3 mg/g. Conclusion The method is suitable for the determination of emodin in Polygonum multiflorum Thunb. The contents of emodin in roots and leaves of Polygonum multiflorum Thunb. from Zhejiang province are different.
