摘要
建立了快速、简单的样品前处理(QuEChERS)用于食用菌中尼古丁含量分析。称取5g新鲜样品(0.5g干样,加入2mL水,混匀)于50mL离心管,加2g NaCl,6gNa2SO4,1mL氨水,加入5mL苯,离心5min。吸取2mL有机相,加入40mg异丙基氨基(PSA)和200mg无水Na2SO4,离心1min。取1μL进样,气相色谱/质谱(GC/MS)定性定量。分别进行同天、不同天和双人测定尼古丁重复性实验,加标回收率实验。经测定,样品的加标回收率大于83%,相对标准偏差小于8%。
A fast and easy to handle sample preparation procedure(QuChERS) for nicotine in mushrooms was developed.A small amount(5 g fresh sample or 0.5 g dried sample blended with 2 mL water) sample was mixed with 2 g NaCl,6 g Na2SO4,1 mL ammonium and 5 mL benzene.The mixture was shaken vigorously and centrifuged for 5 min.Then 2 mL of the supernatant was transferred into a plastic microcentrifuge safelock tube,which contained 40 mg of a mixture of PSA and 200 mg anhydrous sodium sulfate.After centrifuging for another 1 min,the supernatant was analysed by gas chromatography-mass spectrometry(GC-MS).The recoveries are 83% with relative standard deviations 8%.
